Identification and Isolation of the Specific Allergens from Mutton by Western Blot, Chromatography and MALDI-TOF/TOF-MS

ABSTRACT

To study the specific allergen in mutton, soluble protein from mutton was extracted, and separated by SDS-PAGE. With the positive and negative sera of rats, the special allergen was identified with the Western Blot, showing that the 12kDa was the specific allergen. The protein of 12kDa was then analyzed by ion exchange and gel filtration chromatography, and the MALDI-TOF/TOF-MS search on the Internet. The results indicate that the protein was diamine- N-acetyltransferase.
